Production and Characterization of Extracellular L-Asparaginase from Microorganism

Abstract
The regulation for production of L-asparaginase in Candida utilis was investigated. Among tested nitrogen sources, peptone was best for the enzyme production while readily metabolizable nitrogen sources such as various amino acids and ammonium sulfate repressed the enzyme synthesis. The enzyme synthesis was also inhibited when both peptone and readily metabolizable nitrogen sources were used as nitrogen sources. The repression of the enzyme synthesis was also observed when L-asparagine was added immediately prior to the synthesis of the enzyme. L-Asparagine and ammonium sulfate were most effective for repression of L-aspraginase in Candida utilis. Therefore it seems likely that L-asparaginase production in Candida utilis is regulated by nitrogen catabolite repression.
